Abstract: The present invention relates to a synergistic nano-biostimulant composition comprising an Ascophyllum nodosum extract, nano-chitosan, and free-form Lproline, and to a low-temperature ultrasound-assisted process for preparation thereof. The Ascophyllum nodosum extract is obtained by ultrasound-assisted 10 extraction at a frequency of 25–40 kHz, at a temperature maintained below 28°C, at a pH of 6.0–7.0, and for a duration of 30–60 minutes, thereby preserving thermolabile bioactive constituents. The composition comprises 40–60% v/v of the Ascophyllum nodosum extract, 0.5–2% w/v nano-chitosan, and 1–5% w/v free-form L-proline. The nano- chitosan has a mean particle size of 60–90 nm, a polydispersity 15 index of less than 0.30, a zeta potential of +25 to +40 mV, and forms an electrostatically complexed nano-system with the seaweed extract, as confirmed by FTIR analysis. The composition provides a synergistic abiotic-stress mitigation effect, exhibiting an Abiotic Stress Tolerance Index (ASTI) improvement of 31.4%, a Synergy Index of 0.87, and a grain-yield increase of 40.1% under heat-stress 20 conditions as compared with untreated control. The composition is useful for enhancing plant growth and tolerance to abiotic stress in agricultural applications, including foliar and related crop- treatment uses. The formulation further exhibits storage stability for 18–24 months under refrigerated conditions.
