Abstract: The present invention relates to the development of a gastroretentive floating tablet formulation of Ethamsylate for controlled drug delivery and prolonged gastric residence time. The formulation comprises hydrophilic and hydrophobic polymers including hydroxypropyl methylcellulose (HPMC), beeswax, and ethyl cellulose, prepared by a melt granulation technique. Sodium bicarbonate is incorporated as a gas-generating agent to impart buoyancy to the dosage form. Preformulation studies including infrared spectroscopy, melting point analysis, solubility studies, and drug-excipient compatibility confirmed the suitability of the drug for formulation development. A factorial design approach was employed to optimize critical formulation variables affecting floating behavior and drug release characteristics. The prepared tablets exhibited satisfactory physicochemical properties including hardness, bulk density, angle of repose, and weight variation. UV spectrophotometric analysis demonstrated linearity in the concentration range of 5–65 µg/ml with a correlation coefficient of 0.998. The formulation provides sustained drug release and enhanced gastric retention for improved therapeutic efficacy.
