Abstract: The present invention relates to a novel thermoresponsive pulp capping composition, designated as REGEN-CAP, developed for enhanced dentin regeneration and vital pulp therapy. The composition is formulated as a powder—liquid system, wherein the powder comprises Poloxamer 407, tricalcium silicate, amorphous calcium phosphate, nanohydroxyapatite, bioactive glass (45S5), and chitosan, and the liquid component consists of sterile saline. Upon mixing, the formulation exhibits thermoresponsive sol—gel transition at body temperature, allowing easy handling and precise placement in the prepared cavity. The presence of Poloxamer 407 acts as a delivery matrix, significantly enhancing the bioavailability of bioactive ions and enabling controlled and sustained release of calcium silicate-derived ions. The synergistic combination of calcium-releasing and bioactive components promotes mineral nucleation, odontoblastic differentiation, and reparative dentin bridge formation. Additionally, the incorporation of chitosan imparts antimicrobial and bioadhesive properties, while bioactive glass facilitates the formation of a hydroxycarbonate apatite layer. The developed material demonstrates improved sealing ability, reduced microleakage, and superior biological response compared to conventional pulp capping agents. REGEN-CAP is suitable for applications including direct pulp capping, indirect pulp capping, and regenerative endodontic procedures, providing a promising advancement in minimally invasive dental therapeutics.
