Abstract: The present invention provides a 3D bioprintable, biocompatible and stable hydrogel-based formulation that facilitates sustained release of therapeutic molecules for the management of neurodegeneration. The hydrogel-based formulation comprises of an engineered cell as a bioactive material that includes a mammalian cell transfected with a vector encoding a gene of a biomolecule, a selection marker and a promoter sequence, encapsulated in a hydrogel scaffold fabricated with a polymeric ink that releases the biomolecule, thus ensuring the prolonged survival and ensuring delivery of the biomolecule through the blood-brain-barrier. Therefore, the present invention provides a construct with desirable geometry that allows diffusion of nutrients and facilitates blood vessel growth that support the viability of the engineered cells, thus providing efficient management of neurodegeneration.
