Abstract: The present invention relates to a hydroalcoholic flower extract of Sphagneticola trilobata (L.) Pruski and a process for its preparation, exhibiting significant anti-inflammatory and antidiabetic activities. The extract is prepared by Soxhlet extraction of dried and powdered flowers using 70% ethanol at a temperature of 60–70°C for 8–12 hours. The process yields approximately 41.21 g of extract from 500 g of flower powder, corresponding to an extraction yield of 8.24% (w/w). Phytochemical screening revealed the presence of flavonoids, glycosides, tannins, triterpenes, steroids, carbohydrates, and reducing sugars, with flavonoids being the predominant constituents. The anti-inflammatory activity was evaluated in LPS-stimulated RAW 264.7 murine macrophage cells, wherein the extract demonstrated concentration-dependent inhibition of nitric oxide production, achieving 69.90% inhibition at 100 µg/mL. The antidiabetic activity was assessed in HepG2 cells using a glucose utilization assay, wherein the extract enhanced glucose utilization in a concentration-dependent manner and exhibited 166.19 ± 3.86% glucose utilization at 100 µg/mL compared to untreated control cells. The invention further provides pharmaceutical, nutraceutical, and herbal compositions comprising the extract for use in the management of inflammatory disorders, diabetes mellitus, and associated metabolic conditions.
