Abstract: A glucose-responsive pharmaceutical hydrogel for regulated insulin delivery. The system comprises a hydrogel construct (101), defining a three-dimensional, cross-linked polymeric matrix configured to encapsulate insulin-secreting biological units (102). A glucose-responsive sensing matrix (103), comprising a synthetic glucose-binding functional group, is integrated within the polymeric matrix. A permeability-modulation module (104) is operatively coupled to the glucose-responsive sensing matrix (103) and regulates diffusion of insulin through the hydrogel construct (101). An immune-interaction attenuation layer (105) is disposed on an outer surface of the hydrogel construct (101) and comprises engineered non-naturally occurring surface ligands configured to reduce immune-cell mediated recognition. An increase in glucose concentration triggers a reversible structural change in the hydrogel construct (101), thereby enhancing insulin diffusion in a glucose-dependent manner. The invention further provides a method (200) for synthesizing the hydrogel with controlled size, composition, and functional integration of the modules.
