Abstract: The invention relates to an electrochemical biosensor for cardiac troponin I detection. The biosensor comprises a disposable Carbon Screen-Printed Paper Electrode on a 220 GSM cellulose paper substrate in a two-electrode arrangement with a carbon working electrode, a carbon counter electrode, conductive carbon tracks, and contact pads. A borophene layer is disposed on the carbon working electrode, anti-CTnI is immobilized thereon through EDC and NHS mediated coupling, and a 1% BSA blocking layer forms a BSA/anti-CTnI/borophene/CSPPE immunoelectrode. Upon electrochemical interrogation in 5 mM methylene blue in phosphate-buffered saline, CTnI binding alters charge-transfer behavior, enabling detection over 0.01 to 50 ng/mL with a limit of detection of 0.001 ng/mL in buffer and CTnI-spiked human serum.
