Abstract: ABSTRACT OF THE INVENTION Title: A Chronomodulated Pulsatile Pulsincap Drug Delivery System Comprising Tramadol Hydrochloride for Circadian Management of Arthritic Pain The present invention relates to a chronomodulated pulsatile drug delivery system designed to synchronize Tramadol release with biological rhythms for circadian management of arthritic pain. The system comprises drug-loaded microcapsules (100) with Tramadol HCl (110) and Eudragit polymers (120); a modified pulsincap device (200) with cross-linked capsule body (210), HPMC hydrogel plug (220) for 2-3 hour lag time, and enteric coating (230) with cellulose acetate phthalate; chronotherapeutic release mechanism (300); circadian alignment (400) for bedtime administration with early morning release; and therapeutic benefits (500). The invention provides reduced dosing frequency, enhanced patient compliance, and optimized therapeutic efficacy by synchronizing drug release with peak arthritic pain patterns.
