Abstract: The present disclosure proposes a composition (100) of dual-metal ionically crosslinked alginate-glycerol hydrogel comprising a sodium alginate matrix (102), glycerol (104), zinc ions (106), and a secondary metal ion (108) selected from cobalt ions and manganese ions. The zinc ions (106) and the secondary metal ion (108) ionically coordinate with carboxyl groups of the sodium alginate matrix (102) through ionotropic gelation to form a single integrated dual-metal crosslinked hydrogel network. The glycerol (104) forms hydrogen-bond-mediated interactions with hydroxyl and carboxyl groups of the sodium alginate matrix (102). The composition (100) exhibits enhanced antibiofilm activity against Staphylococcus aureus, controlled swelling, structural integrity, mammalian-cell cytocompatibility, and hemocompatibility. The hydrogel further exhibits an amorphous structure and uniform zinc loading, and includes alginate-glycerol-zinc-cobalt and alginate-glycerol-zinc-manganese formulations.
