Abstract: ABSTRACT A CONSTRUCT AND METHOD FOR ENHANCING IRON CONTENT AND IMPROVING ROOT GROWTH IN PLANTS The present invention generally relates to the fields of plant molecular biology, plant physiology, and agricultural biotechnology, and more particularly relates to a system and method for modulating iron deficiency responses, iron homeostasis, rhizosphere acidification, and root growth in plants through PROPEP2-Pep2-PEPR2 signaling. The invention provides construct and methods for manipulating PEP2 expression, either by overexpressing PROPEP2 (the precursor form of PEP2) or by applying synthetic Pep2 peptides. The invention also encompasses the use of transgenic plants with modified PEP2 expression, such as plants with reduced PEP2 function (e.g., pep2 mutants) or plants overexpressing PROPEP2 under constitutive promoters. Further, CRISPR- engineered plants with targeted modification of the PROPEP2 gene exhibit enhanced iron accumulation, higher rhizosphere acidification and improved root growth under iron (Fe)-limiting conditions, thereby providing improved tolerance to iron deficiency stress and enhanced nutrient acquisition efficiency in plants.
