Abstract: [068] A CRISPR-based gene editing delivery system for treating genetic disorders and a method thereof are disclosed. The system comprises a targeting module configured to selectively bind diseased cells, a carrier module adapted to encapsulate a CRISPR-based gene editing composition, a controlled release module for intracellular release, and a monitoring and regulation module configured to reduce off-target effects. The CRISPR-based gene editing composition includes at least one guide RNA and at least one programmable nuclease selected from Cas9, Cas12, Cas13, base editors, prime editors, or variants thereof for modifying a target genetic sequence associated with a genetic disorder. The carrier module may comprise lipid nanoparticles, polymeric nanoparticles, viral vectors, extracellular vesicles, or hybrid nanocarriers enabling targeted in vivo or ex vivo delivery. The disclosed method includes identifying a target gene, preparing and encapsulating the gene editing composition, administering the delivery system, and editing the target genetic sequence to correct or regulate disease-associated genes for therapeutic treatment of genetic disorders.
