Abstract: A dual-stimuli responsive nanocarrier system with sequential activation for targeted drug delivery is disclosed. The system comprises a nanocarrier core containing at least one therapeutic payload, an inner stimulus-responsive layer associated with the core, and an outer shielding layer encapsulating the inner layer. The outer shielding layer is configured to undergo degradation, transformation, or removal upon exposure to a first biological stimulus, thereby enabling subsequent exposure of the inner stimulus-responsive layer to a second biological stimulus. The outer shielding layer substantially restricts interaction between the inner layer and the second biological stimulus prior to its degradation. The inner stimulus-responsive layer is configured to release the therapeutic payload only after sequential exposure to the first biological stimulus followed by the second biological stimulus, thereby enabling controlled, site-specific, and targeted drug delivery.
