Abstract: ABSTRACT Disclosed herein is a fluorescence-enhanced Feulgen micronucleus diagnostic method (100), the method (100) comprises collecting (102) exfoliated oral epithelial cells from a buccal mucosal surface. The method also includes fixing (104) the collected cells in ethanol. The method also includes subjecting (106) the fixed cells to controlled acid hydrolysis. The method also includes contacting (108) the hydrolyzed cells with a Schiff reagent to form a DNA-specific Schiff–DNA complex. The method also includes visualizing (110) the Schiff–DNA complex using fluorescence microscopy. The method also includes quantifying (112) a micronucleus frequency by counting micronuclei per 1000 intact epithelial cells. The method also includes classifying (114) the sample as oral leukoplakia when the quantified micronucleus frequency is equal to or greater than 3 micronuclei per 1000 epithelial cells, and classifying the sample as normal mucosa when the micronucleus frequency is less than 3 micronuclei per 1000 epithelial cells.
