MUMBAI, India, July 31 -- Intellectual Property India has published a patent application (202411102251 A) filed by National Institute Of Pharmaceutical Education And Research Niper on December 23, 2024, for A Green Process For The Preparation Of Efaproxiral Intermediate.

Abstract: The present disclosure relates a green process for preparing an intermediate compound of Efaproxiral represented by formula IV. The preparation method involves the generation of 4-hydroxy phenacetyl radical from 4-hydroxy phenyl-acetohydrazide compound of formula II under the influence of a green oxidant & additive, which ultimately reacts with the amine source, 3,5-dimethyl aniline of formula III to afford the intermediate compound, N-(3,5-dimethylphenyl)-2-(4-hydroxyphenyl) acetamide represented by formula IV in high yield and purity. The preparation process of the present invention avoids precious metal as a catalyst, and accordingly, the cost is low, the post-treatment is simple, the product has a high yield of 95% and HPLC purity of 99%. The prepared intermediate can be effectively used for the synthesis of Efaproxiral and is suitable for its scaled-up industrial production.
