Abstract: Disclosed is a logic-gated multi-layer microencapsulation system (100) for controlled and conditional drug release. The system (100) comprises an inner core (102) encapsulating at least one active pharmaceutical ingredient, an intermediate logic layer (104) surrounding the inner core (102), optionally comprising a time-delay sub- layer (108), and an outer pH-sensitive shell (106) surrounding the intermediate logic layer (104). The outer pH-sensitive shell (106) dissolves, erodes, swells, or becomes permeable at a target pH range. The intermediate logic layer (104) responds to a secondary physiological trigger independent of pH, including enzyme activity, ionic strength, redox condition including reactive oxygen species level, or a combination thereof. Release from the inner core (102) occurs only when both layers are dissolved, eroded, swollen, activated, degraded, disrupted, or rendered permeable by corresponding trigger conditions, thereby providing a material-based AND-gate release mechanism. An optional time-delay sub-layer (108) introduces predetermined release delay.
