Abstract: The present invention discloses a method for biosynthetic fucoidan derivative synthesis and neuropharmacological evaluation for neuroprotection against neurodegenerative diseases. Fucoidan monomer is isolated from Sargassum marginatum through ion exchange chromatography, mild acid hydrolysis, and gel permeation chromatography. Acetylated, benzoylated, and phosphorylated derivatives are synthesized at C3 and C4 hydroxyl positions and confirmed by UV, ATR- FTIR, 1HNMR, and mass spectroscopy. In-silico docking against Acetylcholinesterase, beta-secretase 1, and Glycogen synthase kinase-3beta with MM-GB/SA binding free energy analysis identifies phosphorylated fucoidan as the optimal derivative. In-vitro evaluation in Neuro-2A cells demonstrates superior neuroprotective efficacy through restored cell viability, acetylcholinesterase inhibition, dopamine restoration, enhanced antioxidant activity, preserved mitochondrial integrity, suppressed apoptosis, and upregulated BDNF, Nrf2, and Bcl-2 expression, providing a promising marine-derived lead candidate for neurodegenerative disease management.
