Abstract: A method (100) for detecting circulating tumor cells (CTCs) in blood is disclosed. The method (100) comprising steps of obtaining a peripheral blood sample from a subject; isolating peripheral blood mononuclear cells (PBMCs) from the peripheral blood sample using density-gradient centrifugation and fixing the PBMCs by 4% PFA; contacting and tagging the isolated PBMCs with fluorochrome-conjugated antibodies comprising an epithelial marker EpCAM, a mesenchymal marker N-cadherin, and a stem-cell marker ALDH1; subjecting the antibody-treated PBMCs to analysis using flow cytometer comprising multiple laser sources; exciting the fluorochrome-conjugated antibodies using the multiple laser sources of the flow cytometer; detecting cells exhibiting expression of EpCAM, N-cadherin, and ALDH1 to identify CTCs present in the blood; calibrating detection sensitivity using a spike-in validation framework comprising introduction of known numbers of tumor cells into peripheral blood samples at multiple dilution levels; identifying CTC subpopulations exhibiting epithelial, mesenchymal, or stem-cell-associated phenotypes associated with metastatic competence.
