Abstract: ABSTRACT Disclosed herein is a method (100) for formulation and evaluation of rutin-loaded glycerosomes for enhanced wound healing applications comprises selecting (102) rutin as an active pharmaceutical ingredient, followed by conducting preformulation tests (104) to assess physicochemical characteristics and compatibility. The method further includes formulating rutin-loaded glycerosomes by dissolving the rutin, phospholipid, and cholesterol in an organic solvent system (106). The formulation is optimized (108) using a Central Composite Design by varying key formulation variables. The optimized glycerosomes are evaluated (110) for vesicle characteristics and drug entrapment efficiency, incorporated (114) into a polymeric gel base prepared (112) to obtain a uniform glycerosomal gel, and evaluated (116) for physicochemical properties and in vitro drug release. The method further includes conducting in vivo wound healing studies (118) and performing stability testing (120) under predetermined storage conditions to assess long-term stability and drug retention.
