Abstract: [061] A mucoadhesive buccal film for rapid systemic delivery of pharmaceutical agents and a method of administration thereof are disclosed. The mucoadhesive buccal film comprises a polymeric film-forming matrix including at least one mucoadhesive polymer, one or more active pharmaceutical ingredients, at least one plasticizer, and one or more permeation-enhancing agents configured to facilitate transmucosal absorption through a buccal mucosal surface. The buccal film is configured to adhere to the buccal mucosa and release the pharmaceutical agents in a controlled or rapid manner for systemic delivery while bypassing first-pass hepatic metabolism. The method includes preparing the film-forming composition, casting or molding the composition into a buccal film, drying the film, and administering the film onto the buccal mucosa of a subject for rapid absorption. The disclosed formulation improves bioavailability, onset of action, patient compliance, and ease of administration for diverse therapeutic applications.
