Abstract: A thermoacoustic-triggered intranasal nanocarrier system (100) for targeted delivery of neuroactive agents to the brain. The system comprises a nanocarrier core (101) encapsulating a neuroactive agent (103) and an acoustic sensitizer (104), a thermosensitive shell (102) surrounding the core, a surface modification layer (105) configured to enhance nasal residence and neural transport, and a thermoacoustic response modulation layer (106) disposed between the core and shell to regulate energy transfer. The system further includes a permeation enhancement layer (107) configured to transiently increase epithelial permeability, a stabilization matrix (108) for maintaining structural integrity during storage and administration, and an acoustic coupling and localization module (109) for efficient transmission and focusing of externally applied acoustic waves. A method (200) of manufacturing the system is also disclosed, enabling controlled formation, stabilization, and formulation of the nanocarrier for intranasal administration and targeted, externally triggered drug release.
