Abstract: The present invention relates to a nanozyme-integrated electrochemical biosensor and method for rapid in vitro detection of Creatine Kinase-MB (CK-MB), a clinically significant cardiac biomarker for acute myocardial infarction diagnosis. Conventional ELISA and immunoassay methods require laboratory infrastructure, trained personnel, cold-chain reagents, and 2–4 hours for results. The invention provides a biosensor comprising a conductive electrode substrate functionalized with a catalytic nanozyme layer (Fe3O4, CeO2, or Au nanoparticles with peroxidase-like or oxidase-like activity) covalently conjugated with CK-MB-specific antibodies, enabling amplified electrochemical signal generation upon CK-MB binding. Detection is performed within 5 minutes using differential pulse voltammetry or electrochemical impedance spectroscopy. The system achieves a detection limit of 0.3 ng/mL across a linear range of 1–200 ng/mL in human serum, with superior nanozyme stability over 30 days at ambient conditions, eliminating cold-chain dependency. The invention is applicable for emergency cardiac diagnostics, ambulance-based testing, and point-of-care facilities.
