Abstract: The present invention relates to a novel class of 1,2,3-triazole-linked hydroxycoumarin dimers useful as dual inhibitors of bacterial DNA gyrase and topoisomerase IV and as antibacterial therapeutics. The invention provides compounds represented by Formula I comprising hydroxycoumarin pharmacophores linked through one or more triazole bridges, including pharmaceutically acceptable salts, hydrates, solvates, stereoisomers and prodrugs thereof. The compounds are designed to exhibit dual- target inhibition, multivalent binding interactions and enhanced affinity toward GyrB and ParE binding pockets, thereby overcoming limitations associated with conventional antibacterial agents. The invention further provides processes for synthesis of said compounds through hydroxycoumarin intermediate preparation, propargylation, azide synthesis and coppercatalyzed azide-alkyne cycloaddition (click chemistry) for formation of triazole-linked dimers. Pharmaceutical compositions comprising the compounds and methods for treatment or prevention of bacterial infections, including infections caused by multidrugresistant pathogens, are also disclosed. The invention further encompasses Markush derivatives, molecular hybrid scaffolds and antibacterial lead compounds useful in pharmaceutical development. The disclosed compounds provide a new antibacterial chemotype with potential improved potency, resistance mitigation and broad industrial applicability in anti- infective drug discovery.
