Abstract: ABSTRACT [0027] In this invention, we report the development of an ultrasensitive label-free electrochemical immunosensor for the detection of CRP in human serum and saliva samples. Initially, poly pyrrole integrated with Cu Ni-MOF (PPy @CuNi-MOF) were synthesized using hydrothermal method and subsequently drop-cast onto a glassy carbon electrode (GCE) to fabricate the immunosensor. The structural and morphological properties of the immunosensor were systematically characterized using X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR), field-emission scanning electron microscopy (FE-SEM), energy- dispersive spectroscopy (EDS), and X-ray photoelectron spectroscopy (XPS). Electrochemical performance was evaluated using cyclic voltammetry (CV), differential pulse voltammetry (DPV), and electrochemical impedance spectroscopy (EIS). The developed immunosensor demonstrated a wide linear detection range from 0.00005 to 500 ng/mL, along with excellent sensitivity, reproducibility, repeatability, and long-term stability. Selectivity studies confirmed negligible interference from common biomolecules and inorganic ions. Furthermore, the practical applicability of the sensor was validated using human serum and saliva samples for the detection of CRP.
