Abstract: The present invention relates to a novel polyherbal formulation exhibiting anti-diabetic and anti-acidity activity, comprising an ethanolic extract of leaves of six medicinal plants, namely Moringa oleifera, Coriandrum sativum, Mentha arvensis, Cynodon dactylon, Mangifera indica and Ficus religiosa, combined in substantially equal proportions. The extract is prepared by Soxhlet extraction using ethanol as solvent at 40°C–60°C for 18–22 hours, and is characterized by FTIR spectroscopy confirming the presence of hydroxyl, carbonyl, aromatic and amine functional groups indicative of phenolic and flavonoid phytoconstituents. Biological evaluation in an alloxan-induced zebrafish (Danio rerio) model at doses of 200 mg, 400 mg and 600 mg demonstrates a dose- dependent reduction in blood glucose levels without significant toxicity, confirming the anti-diabetic potential of the formulation, while its antioxidant phytoconstituents further support gastroprotective/anti-acidity activity. The formulation is natural, safe, non-toxic and suitable for further pharmaceutical development into various oral dosage forms
