Abstract: The present invention relates to a polymeric transdermal drug delivery system comprising Remogliflozin Etabonate for sustained management of diabetes mellitus. The transdermal patch comprises a combination of hydroxypropyl methylcellulose, Eudragit RLPO, Eudragit RSPO, a polyvinyl alcohol backing membrane, and a plasticizer for regulating drug release and maintaining structural integrity. The patch is prepared by a solvent casting and evaporation process to obtain a flexible film suitable for prolonged skin application. Formulation variables are optimized using a Box–Behnken experimental design to achieve controlled physicochemical characteristics and sustained drug release. The developed system enables transdermal permeation of Remogliflozin Etabonate over an extended period. In vivo evaluation in streptozotocin-induced diabetic animals demonstrates effective glycemic control and improvement in associated biochemical parameters. The invention provides a sustained-release transdermal therapeutic system for long-term antidiabetic treatment.
