Abstract: The present invention discloses a novel small molecule bioconjugate comprising folic acid conjugated with curcumin for targeted delivery to triple-negative breast cancer (TNBC) cells. The bioconjugate is designed to selectively bind folate receptor-a (FRa) overexpressed on TNBC cells, facilitating receptor-mediated internalization and intracellular release of curcumin. The conjugation is achieved via a pH-sensitive ester linkage using Steglich esterification, and the compound is characterized by spectroscopic and analytical techniques. The invention further encompasses molecular docking studies, in vitro anticancer and apoptosis assays, and chemo sensitization studies, demonstrating enhanced intracellular accumulation, cytotoxicity, and therapeutic efficacy compared to unconjugated curcumin. This small molecule bioconjugate provides a simple, targeted, and effective strategy for TNBC therapy and potentially other FRa-overexpressing cancers.
