Abstract: A pharmaceutical composition for controlled gastric retention is disclosed. The composition comprises a drug containing core, hydrophilic swelling matrix, gas generating component, gel strengthening polymeric phase, and release modifying excipients. Upon contact with gastric fluid, the composition hydrates, expands, generates entrapped gas, and forms a cohesive gel barrier that supports gastric residence and controlled release of an active pharmaceutical ingredient. The system is suitable for drugs benefiting from prolonged gastric or upper intestinal exposure and can be manufactured using conventional pharmaceutical processes.
