Abstract: The present invention relates to a polyherbal ayurvedic vaginal suppository formulation for localized vaginal administration. Each two grams suppository comprises equal quantities of Vata (Ficus bengalensis), Udumbar (Ficus glomerata), Plaksha (Ficus infectoria), Ashwatha (Ficus religiosa), Parisha (Thespesia populnea), Nimba (Azadirachta indica), Haridra (Curcuma longa) and Yashtimadhu (Glycyrrhiza glabra), wherein each herb is present at 62.5 mg and the total herbal drug load is 500 mg. The formulation further comprises chitosan in the range of 100-150 mg as a mucoadhesive retention-enhancing component and cocoa butter quantum sufficient to make a two grams suppository. The preparation process includes drying, powdering, #80 sieving, equal-proportion blending, melting cocoa butter at 30-35 degrees Celsius, preparing chitosan gel in 1% acetic acid, incorporating the herbal mixture and gel, moulding and cooling. The formulation provides synergistic astringent, antimicrobial, anti-inflammatory, soothing and mucosal healing action.
