Abstract: ABSTRACT Developing glyphosate insensitive Class I EPSP synthase for herbicide tolerant transgenic crop plants for higher crop productivity The present invention relates to genetically modified plants, particularly monocotyledonous crops such as rice, exhibiting enhanced agronomic performance, including increased grain yield, robust glyphosate tolerance, improved lodging resistance, optimized plant architecture, and stress resilience. The invention provides a recombinant nucleic acid encoding a glyphosate-insensitive Class I 5-enolpyruvylshikimate-3-phosphate (EPSP) synthase, preferably of cyanobacterial origin, targeted to plastids. Expression of this enzyme relieves the primary flux-controlling bottleneck in the shikimate pathway, leading to the overaccumulation of aromatic amino acids without disrupting endogenous metabolic regulation. Elevated tryptophan enhances auxin biosynthesis, improving panicle development and grain filling, while elevated phenylalanine and tyrosine enhance lignification and secondary metabolite production, conferring lodging resistance and stress tolerance. Transgenic plants expressing the enzyme demonstrate robust tolerance to glyphosate and significant increases in yield compared to non-transgenic controls. The invention further provides methods for producing such plants, recombinant constructs, and related compositions.
