Abstract: ABSTRACT A PROCESS FOR ISOLATING BETA-GLUCAN FROM MARINE ENDOPHYTIC DEBARYOMYCES SPECIES ASSOCIATED WITH GRACILARIA EDULIS The present disclosure provides a process (100) for isolating beta-glucan from a marine endophytic Debaryomyces species. The process (100) comprises collecting a macroalgal sample comprising Gracilaria edulis from a marine coastal region and transporting the macroalgal sample under sterile conditions (102). The process (100) further comprises surface sterilizing the macroalgal sample (206) and isolating an endophytic yeast from the macroalgal sample by inoculating sterilized tissue onto culture media supplemented with an antibiotic (208). The process (100) comprises cultivating the endophytic yeast in a liquid culture medium to obtain yeast biomass (212) and extracting crude beta-glucan (314) from the yeast biomass by subjecting the yeast biomass to a heat treatment (304) followed by an alkaline lysis (306) and an acid treatment (310). The process (100) further comprises purifying the crude beta-glucan (314) through a multi-step purification cascade (108) comprising alkaline re-solubilization (402), deproteinization (406, 408), delipidation (410, 412, 414), and dialysis (416) to obtain purified beta-glucan (600).
