Abstract: An enzyme-free nucleic acid stabilizing composition (100) for protected oral delivery of nucleic acids. The composition (100) comprises a supramolecular scaffold (102) formed from a pH-responsive copolymer and a polyol, the scaffold (102) defining a three- dimensional nano-porous network configured to physically entrap a nucleic acid payload (104). A mucoadhesive stabilization matrix (106) is disposed in association with the supramolecular scaffold (102) to enhance mucosal retention and improve localized absorption. A protective enteric modulation layer (108) is provided to maintain structural integrity of the supramolecular scaffold (102) under acidic gastric conditions and to enable release at intestinal pH. An osmotic hydration regulator (110) is incorporated to maintain hydration and preserve conformational stability of the nucleic acid payload (104). The composition exhibits thermoreversible phase transition behaviour, enabling efficient encapsulation and controlled release. The invention further provides a method (200) for preparation of the composition suitable for pharmaceutical and biotechnological applications.
