Abstract: The present invention relates to a self-emulsifying nanostructured pharmaceutical composition for enhancing the bioavailability of poorly water-soluble drugs. The composition comprises a poorly soluble active pharmaceutical ingredient, a lipidic solubilizing phase, a pharmaceutically acceptable surfactant, a co-surfactant, a crystallization inhibitor and a solid carrier or adsorbent. Upon contact with gastrointestinal fluid, the composition spontaneously forms nano-sized droplets or a nanostructured dispersion that maintains the active ingredient in a solubilized or supersaturated absorbable form. The lipidic phase improves drug solubilization, the surfactant and co-surfactant facilitate spontaneous emulsification, and the crystallization inhibitor prevents precipitation after dilution. The composition may be provided as a capsule, tablet, powder, granule, pellet or sachet. The invention improves dissolution rate, gastrointestinal dispersion, mucosal availability, absorption and therapeutic consistency of poorly soluble drugs, while providing a scalable and stable pharmaceutical dosage platform.
