Abstract: ABSTRACT A standardized herbo-mineral composition (Mrityunjaya Rasa) and a process for preparation thereof for therapeutic application as an antitumor and immunomodulatory agent The present invention relates to a novel Ayurvedic herbomineral formulation known as Mrityunjaya Rasa, a standardized composition comprising Hirak Bhasma, Mukta Pishti, Rasasindoor, and Nimbu Swarasa, along with a defined carrier system. The invention further discloses a standardized manufacturing process (SMP), pharmacopeial and physicochemical standards derived from multiple batches, and a therapeutic application as an antitumor and immunomodulatory agent. Preclinical evaluation using a HeLa human cervical cancer xenograft in immunodeficient mouse model demonstrates significant antitumor and immunomodulatory activity, mediated through modulation of the tumor microenvironment and reduction of pro-inflammatory cytokine IL-6. Transcriptomic analysis provides mechanistic insight into the formulation’s mode of action, demonstrating modulation of multiple tumor-associated signaling pathways in xenograft tissue along with regulation of the host microenvironment, contributing to control of inflammation and tumor progression. The formulation offers a cost-effective, integrative, and evidence-based therapeutic platform, bridging traditional Ayurvedic knowledge with modern biomedical validation.
