Abstract: [053] A stimuli-responsive nanocarrier system for controlled therapeutic release in tumor microenvironments and method thereof are disclosed. The system comprises a nanocarrier matrix configured to encapsulate one or more therapeutic agents, at least one stimuli- responsive component responsive to tumor-associated stimuli, a targeting moiety configured for selective tumor localization, and a protective surface layer configured to enhance systemic stability. The stimuli-responsive component undergoes physicochemical transformation in response to conditions including acidic pH, enzymatic activity, hypoxia, redox variation, temperature variation, or reactive oxygen species present within tumor microenvironments, thereby enabling localized and controlled release of therapeutic agents. The disclosed method includes encapsulating therapeutic agents, functionalizing the nanocarrier matrix with targeting and stimuli-responsive components, administering the system to a subject, accumulating the system at a tumor site, and triggering controlled release within the tumor microenvironment. The disclosed system improves targeted therapeutic delivery while reducing premature release, systemic toxicity, and off-target effects.
