Abstract: ABSTRACT A SYSTEM AND A METHOD FOR MULTI-ENZYME RESPONSIVE, LOGIC-GATED PRODRUG ACTIVATION AND TARGETED ANTIBIOTIC DELIVERY AGAINST DRUG-RESISTANT BACTERIA The present disclosure relates to a system and method for multi-enzyme responsive, logic-gated prodrug activation and targeted antibiotic delivery aimed at drug- resistant bacteria. The system has a prodrug engineering module that makes a multi-responsive prodrug with a beta-lactam antibiotic core and cleavable linkers. It also has a sensing module that can find biochemical parameters like enzyme activity, pH changes, protease presence, and quorum sensing signals. Finally, it has a processor that can use predefined logical activation conditions to evaluate the parameters it finds. The system also has a pathogen-adaptive modulation module that changes activation thresholds on the fly and a targeted micro-reservoir delivery module that releases drugs in a specific area over time. A safety suppression module stops activation in areas that aren't meant to be activated. The method allows for targeted activation of the antibiotic at infection sites, enhancing therapeutic efficacy, minimizing systemic toxicity, and addressing the shortcomings of traditional single-trigger drug delivery systems.
