Abstract: The invention provides a system and method for two dimensional and three dimensional fluorescence polarization anisotropy imaging using modulated illumination to achieve scattering suppressed, polarization resolved fluorescence measurement. A laser beam is conditioned and imprinted with a periodic intensity pattern by a spatial modulation device, then shaped into a thin light sheet and polarized through a half wave plate and excitation polarizer. The modulated, polarization controlled sheet illuminates a specimen, and emitted fluorescence is spectrally filtered and divided into two optical paths for parallel and perpendicular polarization detection using analyzer polarizers and synchronized cameras. A reconstruction module applies structured illumination reconstruction to isolate modulation preserving fluorescence originating from singly scattered photons, subsequently computing anisotropy from the reconstructed polarization components. An axial scanning mechanism enables depth resolved acquisition for volumetric anisotropy imaging. The invention provides enhanced anisotropy accuracy and image contrast in turbid or optically dense samples by mitigating scattering induced depolarization.
