The application for the patent was published on October 30, 2026, under issue no. 44/2026.

Abstract: A THERMOSENSITIVE IN-SITU GEL COMPRISING TETRANDRINE LOADED PLGA NANOPARTICLES ABSTRACT The present invention provides a thermosensitive in-situ gel composition for intranasal delivery of tetrandrine, comprising tetrandrine-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles dispersed in an aqueous thermosensitive gel matrix comprising Poloxamer 407 and a mucoadhesive polymer. The formulation is administered in a relatively low- viscosity sol state and undergoes a sol-to-gel transition upon exposure to nasal physiological temperature, thereby increasing viscosity and providing prolonged retention at the nasal mucosal surface. The PLGA nanoparticles provide encapsulation and controlled release of tetrandrine, while the thermosensitive and mucoadhesive matrix provides temperature-responsive gelation and nasal retention. The formulation is configured for nose-to-brain delivery of tetrandrine through nasal transport pathways, including olfactory and trigeminal pathways, and is useful for delivery of tetrandrine for the treatment of glioblastoma.
