Abstract: The present invention relates to a validated reverse-phase high-performance liquid chromatographic method for simultaneous estimation of Ixempra and capecitabine. The method employs a C18 stationary phase and a mobile phase comprising phosphate buffer having a pH of about 7.4 and methanol in a volumetric ratio of about 85:15. A sample comprising Ixempra and capecitabine is introduced into the chromatographic system, preferably at an injection volume of about 20 µL. Ixempra is detected at approximately 254 nm and capecitabine is detected at approximately 254 nm. Under the investigated chromatographic conditions, Ixempra and capecitabine exhibit retention times of approximately 2.2 minutes and 4.8 minutes, respectively. The method demonstrates linearity over approximately 80–220 µg/mL for both analytes, with calculated coefficients of determination of approximately 0.9941 for Ixempra and 0.9963 for capecitabine. The method further demonstrates satisfactory precision and robustness under investigated conditions and is useful for simultaneous quantitative determination and routine quality-control analysis of pharmaceutical compositions comprising Ixempra and capecitabine.
