Abstract: ABSTRACT An engineered fast-elongating Taq DNA polymerase with high blood resistance and allele-specific amplification. The present invention relates to thermostable DNA polymerases exhibiting enhanced resistance to inhibitory substances present in whole blood and related biological matrices. In particular, double-mutant and triple-mutant forms of Thermus aquaticus (Taq) DNA polymerase have been developed that remain catalytically active in reaction mixtures whole blood or equivalent blood fractions, have fast elongation speed and also show allele specificity. These mutant enzymes enable efficient and specific polymerase chain reaction (PCR) amplification of nucleic acid targets directly from unprocessed blood samples, eliminating the need for DNA extraction or purification. The invention further provides compositions, methods, and diagnostic kits utilizing such polymerases for rapid, sensitive, and economical amplification of nucleic acids in clinical, forensic, and analytical applications. The improved inhibitor resistance, catalytic efficiency, and kinetic performance of the disclosed polymerases substantially enhance the speed, reliability, and practicality of PCR-based assays involving complex biological specimens.
