Abstract: [061] The present disclosure relates to an engineered probiotic composition for modulating gut-brain axis disorders and methods of use thereof. The composition comprises at least one genetically modified probiotic microorganism configured to survive gastrointestinal transit and produce one or more neuroactive biomolecules, including neurotransmitters, neuromodulators, anti- inflammatory agents, or short-chain fatty acids. The engineered probiotic microorganism may be formulated with pharmaceutically acceptable carriers, stabilizers, or encapsulating matrices for targeted intestinal delivery and enhanced viability. Upon administration, the composition modulates bidirectional signaling between the gastrointestinal system and the central nervous system for alleviating, preventing, reducing, or managing disorders associated with gut-brain dysregulation. The disclosed composition and method may be useful in treatment or management of anxiety disorders, depressive disorders, neuroinflammatory conditions, autism spectrum disorders, irritable bowel syndrome, cognitive dysfunction, and related neurological or gastrointestinal disorders.
