Abstract: An enzyme responsive drug release matrix system is disclosed. The system comprises a polymeric carrier framework, enzyme cleavable linker domains, drug containing reservoirs, a permeability regulating phase, and optional stabilizing and barrier components. The matrix retains an active pharmaceutical ingredient under non triggered conditions and increases release upon exposure to a selected enzyme. Enzymatic cleavage relaxes the carrier framework, opens diffusion pathways, and permits controlled liberation from reservoirs. The system is adaptable to oral, mucosal, topical, injectable, implantable, or multiparticulate dosage forms.
