Abstract: ABSTRACT: “AN IMPROVED PROCESS FOR PURIFICATION OF RECOMBINANT PROTEINS” The present invention discloses an improved purification process for recombinant fusion or chimeric protein of meningococcal proteins. The improved purification includes purifying a tagged recombinant protein to obtain purified tagged protein by using at least two chromatographic columns connected in series; removing the tag from the purified tagged protein to obtain a recombinant fusion/chimeric protein; and purifying the recombinant fusion/chimeric protein obtaining after tag removal using the anion exchange chromatography column followed by the metal affinity chromatography column to obtain a sterile filtered purified protein. The improved process of purification of tagged protein is carried out by using anion exchange chromatography column connected in series with metal affinity chromatography column. with the improved process results in significant reduction in HCPs and bacterial endotoxins content. The present invention provides an efficient platform for the purification of factor H binding protein-Porin A (fHbp-PorA) fusion proteins used in vaccine against Meningococcal diseases.
