Abstract: The invention provides an integrated microfluidic cartridge (100) for performing extraction-free direct reverse transcription polymerase chain reaction (RT-PCR) comprises an integrated microfluidic cartridge housing (110), a sample inlet port (120), a microfluidic channel network (130), a lysis chamber (140), reagent storage chambers (150), a reagent release and mixing mechanism (155), a reverse transcription chamber (160), a PCR amplification chamber (170), one or more on-chip thermal control zones (180), heating elements or a thermal interface layer (180A), an optical detection window (190), a multi-layer substrate structure (200), one or more sealing membranes and valves (210), and cartridge-reader interface features (220). The cartridge enables sample introduction, nucleic acid release, automated reagent rehydration and mixing, reverse transcription and nucleic acid amplification within a sealed disposable platform without a separate nucleic acid extraction procedure.
