Abstract: The invention relates to an oral drug delivery system with enzyme protection for enzyme-sensitive therapeutic agents. The system comprises a therapeutic core, an inner stabilizing matrix, an enzyme-interception layer, a hydration-regulating membrane, and an outer gastrointestinal transit barrier. The outer barrier protects the dosage unit in gastric conditions, while the internal layers regulate hydration and reduce enzymatic degradation during intestinal release. The system supports controlled oral delivery of peptides, proteins, nucleic acids, probiotics, biologics, and labile small molecules.
