Abstract: [063] The present invention relates to an oral lipid-based drug delivery composition for enhancing bioavailability of poorly water- soluble drugs and a method for preparation thereof. The composition comprises at least one poorly water-soluble active pharmaceutical ingredient, one or more lipid excipients, and at least one surfactant configured to promote formation of a lipidic dispersion system upon contact with an aqueous medium. Optional excipients including co-surfactants, stabilizers, polymers, and bioavailability enhancers may also be incorporated. The composition improves solubilization, dissolution, intestinal permeability, and oral absorption of the active pharmaceutical ingredient following oral administration. The invention further provides a preparation method involving formation of a lipid phase, incorporation of surfactants and optional excipients, homogenization or stabilization processing, and conversion into an oral dosage form. The developed formulation enables improved therapeutic efficacy and reduced variability associated with poorly water-soluble drugs.
