Abstract: The present invention discloses a cubosome nanocarrier composition comprising phytantriol as a cubic phase forming lipid and Pluronic F127 as a steric stabilizer, co- loaded with andrographolide and berberine for synergistic antiviral activity against Chikungunya virus. The composition exhibits bicontinuous Pn3m cubic phase symmetry wherein andrographolide partitions within aqueous nanochannels and berberine intercalates within lipid bilayer domains, achieving encapsulation efficiency exceeding 90 percent and drug loading capacity of at least 15%. The invention further provides a top-down high-pressure homogenization process yielding monodisperse nanoparticles of 100-200 nm with polydispersity index below 0.20. The co-encapsulated compounds demonstrate a Combination Index of 0.38 confirming strong synergy, with EC50 of 0.38 micromolar and selectivity index exceeding 60 against CHIKV, representing three-fold improvement over liposomal formulations.
