Abstract: The present invention discloses an Angiopep-2 functionalized, reactive oxygen species responsive, exosome-mimetic nanovesicle encapsulating the selective PERK inhibitor GSK2606414 for targeted treatment of glioblastoma multiforme. The nanovesicle comprises a lipid bilayer of hydrogenated soy phosphatidylcholine, cholesterol, DSPE-PEG2000 and DSPE-PEG2000-Angiopep-2 in which a thioketal-linked cleavable lipid bridge, the synthesis of which is fully exemplified, is incorporated, together with a reconstituted monocyte or macrophage plasma membrane protein fraction.
