Abstract: ABSTRACT TITLE: ANTIMICROBIAL ACTIVITY AND PHYTOTOXICITY OF SILVER NANOPARTICLES SYNTHESIZED FROM NEOSCONA MUKERJII SPIDER WEB SILK PROTEIN The present invention discloses a green synthesis method for silver nanoparticles utilizing alkaline-hydrolyzed silk protein extracted from the web of Neoscona mukerjii spider species as a biological reducing and capping agent. The synthesized spider silk protein-mediated silver nanoparticles (SP- AgNPs) exhibit a surface plasmon resonance peak at 420 nanometers, face-centered cubic crystalline structure, and spherical morphology with sizes ranging from 10 to 50 nanometers. The SP-AgNPs demonstrate significant antimicrobial activity against Escherichia coli and Staphylococcus aureus with minimum inhibitory concentrations of 40 and 50 micrograms per milliliter respectively. Phytotoxicity assessment on Vigna radiata and Triticum aestivum reveals a biphasic concentration- dependent response wherein lower concentrations stimulate seed germination and root elongation while higher concentrations inhibit growth. The invention provides an environmentally sustainable, cost-effective nanoparticle synthesis approach valorizing spider web waste biomaterial for antimicrobial applications with established phytotoxicity safety parameters.
