Abstract: The present invention discloses an apparatus (100) and method for non-invasive preclinical modeling of phosgene-induced cerebral ischemia through the lung-brain axis. The apparatus (100) comprises a polycarbonate exposure chamber (110), a piston assembly (120), a Stryker-flow suction irrigator (130), a nebulizer unit (140), an aerosol delivery conduit (150), a phosgene solution reservoir (160), an airflow regulator (170), and a monitoring and control unit (180). The method employs the apparatus (100) to administer 400 ppm phosgene aerosol for 90 minutes daily over 28 days under normobaric conditions in compliance with OECD test guideline 403, inducing progressive oxidative stress, neuroinflammation, blood-brain barrier disruption, and cognitive impairment in Wistar rats via the lung-brain axis. The invention provides a validated non-invasive preclinical platform with comprehensive multi-modal endpoint evaluation, enabling identification of hypoxia-inducible factor-1 alpha as a novel therapeutic target.
