Abstract: ABSTRACT TITLE: BIOACTIVE-LOADED POLYMERIC TRANSDERMAL COMPOSITION FOR TARGETED TREATMENT OF LOCALIZED TISSUE INFECTIONS The present invention relates to a polymeric transdermal pharmaceutical composition for targeted treatment of localized tissue infections comprising berberine hydrochloride encapsulated within poly(lactic-co-glycolic acid) nanoparticles dispersed in a bioadhesive hydrogel matrix of chitosan and Carbopol. The nanoparticles exhibit mean particle size of 165 nanometres, entrapment efficiency of 86.4 percent and drug loading of 7.8 percent. The composition achieves sustained biphasic release of berberine over 24 hours, enhanced antimicrobial activity with two-fold reduction in minimum inhibitory concentration against Staphylococcus aureus, MRSA, Staphylococcus epidermidis and Escherichia coli compared with free berberine, and 60 to 80 percent biofilm inhibition against staphylococcal biofilms. The formulation demonstrates superior tissue retention, controlled transdermal permeation and skin-compatible physicochemical properties suitable for topical application to infected skin. The invention provides an effective, biodegradable, biocompatible antimicrobial platform addressing limitations of conventional berberine topical preparations.
